Abstract:
This study aims to evaluate side effects of prazosin hydrochloride in fertility of male rats. Study groups were received orally graded dose of the prazosin hydrochloride (20,40, 80) mg/kg for 8-weeks, after end experimental, the treated group show significant decrement in testosterone and luteinizing hormone levels and decrement in sperm count. Also it causes decreased in follicle stimulating hormone level and testicular morphology, Sperm viability and motility. the body weight was decreased in the treated groups compare with control group, and decrease in weight of the tests, seminal vesicles and prostate gland compare with control group. The present study concluded that the prazosin hydrochloride possessed as anti-androgenic and anti spermatogenic affects.

Introduction
Alpha blockers considered as one of the most used therapies for treated chronic hypertensive, it used being at early of 1970, phesition reported that patients might benefit from treatment with this drug (Lee et al., 2008).
The mechanism action of the hypotensive Prazosin hydrochloride is unknown, some investigators reported that it causes decrement in total peripheral resistance and vascular smooth muscle relaxation , a study suggested that the vasodilator effect of prazosin associated to postsynaptic alpha-adrenoceptors blockad ( Frishman and Jorde , 2000).
Prazosin hydrochloride is metabolizing by cytochrome p 450 enzyme which eliminated in urine about 10% ( Hellstrom and Sikka, 2007; Brophy et al., 2010).
Antihypertensive drugs can be used to lower blood pressure (BP). the mechanisms of lowering BP are differentiated from one to other. it can be explanation by the antihypertensive action basis to the following classes; agents of alpha-receptor blocking, agents of beta-receptor blocking, the blockers of calcium channel, angiotensin converting enzyme inhibitors, angiotensin II receptor antagonists and diuretics (Nobile et al., 1997; Neal et al., 2000).
Another study proposed the alfa 1-anderoreceptore closed as effective anti-fertility in male rats. As well as, the anti-fertility effect is related with enhancement in ejaculatory ( Moffat et al., 2005 ) . Prazosin and Tamsulosin interfered with seminal emission in the tail of epididymis cause sperm retention. Also these drugs can either decrease vaginal sperm numbers or cause infertility of an ejaculation (Robaire and Orgebin ,2006). In current study, we attempt to investigate the role of Prazosin hydrochloride on the fertility of albino male rats in terms of sperm motility, number and the morphological changes, testes structure and level of testosterone.
Materials and methods
About 40 male rats weighting ( 300-345) g were used in present study, they housed in a plastic caged under standard environment condition( temperature 22-25 ºC and 12 hr., light-dark cycle) and allowed access to standard laboratory water and feed. They were divided in to four groups (10) animals for each group.
The Prazosin was obtained from (Pfizer lab, Germany) the Prazosin hydrochloride dose (20, 40, 80 mg/kg) were prepared by dissolving (1) gram from Prazosin in (10) ml from distill |
